A PILOT FEASIBILITY STUDY TO INTEGRATE PREP INTO EMERGENCY DEPARTMENTS USING DECISION SUPPORT TOOLS - PROJECT SUMMARY ENDING THE HIV EPIDEMIC IN THE UNITED STATES WILL REQUIRE INNOVATIVE STRATEGIES FOCUSED ON BOTH HIV PREVENTION AND CARE. WHILE PRE-EXPOSURE PROPHYLAXIS (PREP) IS A HIGHLY EFFECTIVE BIOMEDICAL INTERVENTION THAT CAN REDUCE HIV INCIDENCE, UPTAKE HAS BEEN MODEST, PARTICULARLY AMONG GROUPS AT HIGHEST RISK FOR ACQUIRING HIV (E.G., MSM, YOUNG PEOPLE, RACIAL/ETHNIC MINORITIES, AND WOMEN OF COLOR). BARRIERS TO SUCCESSFUL PREP INITIATION INCLUDE STRUCTURAL AND INDIVIDUAL-LEVEL ISSUES AS WELL AS SOCIAL DETERMINANTS OF HEALTH (SDOH). INNOVATIVE STRATEGIES ARE NEEDED TO IMPROVE PREP AWARENESS, ACCESS, AND UPTAKE IN VENUES THAT ROUTINELY CARE FOR VULNERABLE PATIENTS. EMERGENCY DEPARTMENTS (ED) SERVE AS A SAFETY NET FOR UNDERSERVED INDIVIDUALS THAT ARE UNLIKELY TO ACCESS OR ENGAGE IN ROUTINE HEALTH SERVICES. EFFORTS TO SCALE-UP PREP AND EXPAND ITS REACH HAVE INCLUDED ED-BASED PREP PRESCRIBING, YET PREP UPTAKE, LINKAGE TO CARE AND PERSISTENCE HAVE PROVEN CHALLENGING. A FOCUS ON APPLICATION OF EVIDENCE-BASED INTERVENTIONS USING IMPLEMENTATION SCIENCE (IS) TO STRENGTHEN PREP DELIVERY IN THE ED AND URGENT CARES (UC) TO THOSE WHO WILL BENEFIT MOST HAS BEEN LIMITED TO DATE. THIS PROJECT SEEKS TO USE ESTABLISHED IS METHODS TO DEVELOP, VALIDATE, AND PILOT TEST A PATIENT-CENTERED DECISION SUPPORT TOOL (DST) TO PROMOTE PREP PERSISTENCE POST ED/UC CARE. BASED ON THE WHO SDOH FRAMEWORK, WE SEEK TO DEVELOP A DST THAT CAN BE ADMINISTERED IN ED/UC SETTINGS AND WILL ASSIST PREP ELIGIBLE PERSONS TO IDENTIFY THEIR HIV RISK, PROMOTE SELF-EFFICACY, AND PROVIDE SUPPORT FOR SDOH NEEDS IDENTIFIED THROUGH THE TOOL. FIRST, WE WILL USE PILOT DATA, PREVIOUSLY VALIDATED TOOLS, AND EVIDENCE FROM THE ED PREP IMPLEMENTATION LANDSCAPE TO DESIGN A PATIENT- CENTERED PREP DST USING THE CONSOLIDATED FRAMEWORK FOR IMPLEMENTATION RESEARCH. WE WILL THEN REFINE THE DST USING A PARTICIPATORY PROTOTYPING WORKSHOP ENGAGING CURRENT, PRIOR AND POTENTIAL PREP USERS. THE DST WILL BE DESIGNED TO OPTIMIZE PREP INITIATION, ADHERENCE AND PERSISTENCE POST-ED/UC CARE BY IMPROVING HIV RISK ASSESSMENT, SELF-EFFICACY, AND BY IDENTIFYING AND ADDRESSING THE MULTIFACETED MEDICAL AND SOCIAL NEEDS OF THE INDIVIDUAL PATIENT. NEXT, WE WILL DETERMINE THE ACCEPTABILITY OF THE DST USING THE NORMALIZATION PROCESS THEORY ACROSS EDS AND UCS IN THE DC-BALTIMORE REGION. FINALLY, WE WILL CONDUCT PRELIMINARY EFFECTIVENESS TESTING OF THE DST AMONG 120 PREP ELIGIBLE PATIENTS THROUGH A PILOT IMPLEMENTATION STUDY AT TWO EDS AND ONE UC IN BALTIMORE AND DC COMPARING PREP PRESCRIPTION ONLY TO PREP AND THE DST. THE PRIMARY OUTCOME WILL BE 4- MONTH PREP PERSISTENCE; WE WILL ALSO MEASURE PREP LINKAGE TO CARE, INITIATION, PERSISTENCE, AND ADHERENCE AT 1-WEEK, 1- AND 4-MONTHS USING SELF-REPORTED, CLINICAL, AND BIOLOGICAL MEASURES. THIS PROJECT ALIGNS WITH NIH AND EHE PRIORITIES AS IT SEEKS TO IMPROVE THE PREP CONTINUUM OF CARE IN TWO URBAN AREAS WITH HIGH HIV PREVALENCE RATES, REDUCE HEALTH INEQUITIES, AND IMPROVE HEALTH OUTCOMES. SEVERAL CUSTOMERS THAT WOULD BENEFIT FROM THE INCREASED CAPACITY AND REDUNDANCY CREATED BY THE NEW ROUTES WHICH WILL HELP LOWER THE COST OF BANDWIDTH SERVICES (SUB 1 GBPS) IN RURAL AREAS WHERE THE COSTS OF CONNECTIVITY MAKE RATES UNECONOMIC, WHICH INHIBITS FUTURE CONNECTIVITY GROWTH IN THE REGION. THIS PROJECT WILL NOT ONLY INCREASE THE RESILIENCY AND REDUNDANCY OF THE MIDDLE MILE NETWORK, BUT IT WILL ALSO REDUCE COSTS AND SPUR THE MORE RAPID DEPLOYMENT OF LAST MILE SERVICES IN RURAL AREAS. THE PROPOSED PROJECT WILL IMPACT A TOTAL OF 78 COMMUNITIES. ACCORDING TO JUNE 2021 FCC FORM 477 DATA, 11.77% OF THE CENSUS BLOCKS ARE UNSERVED AND 14.05% ARE UNDERSERVED. IN ADDITION, 42 WIRELINE PROVIDERS, 19 WIRELESS PROVIDERS, AND 876 TOWERS WILL BENEFIT FROM THE NEW NETWORK. 16 COMMUNITY ANCHOR INSTITUTIONS ARE WITHIN 1,000 FEET OF THE PROPOSED NETWORK (5 HIGHER EDUCATION, 6 SCHOOLS, AND 5 HEALTHCARE FACILITIES). SEVERAL TRIBAL NATIONS BENEFITING FROM THIS ROUTE INCLUDE THE PEORIA TRIBE, CHEROKEE NATION, QUAPUAW NATION, SHAWNEE NATION, AND THE MIAMI TRIBE. THIS ROUTE WILL SUPPORT RANCHES DESIRING A HIGHER GRADE OF FIBER CONNECTIVITY TO CONNECT THE MANY NEW BROADBAND DEVICES USED IN MODERN AGRICULTURE PRACTICES. THE PROPOSED NETWORK WILL ENABLE THE CONVERSION OF CELLULAR TOWERS FROM MICROWAVE TECHNOLOGY TO FIBER OPTICS, WHICH WOULD ALLOW EXISTING AND NEW CUSTOMERS ACCESS TO BANDWIDTH IN EXCESS OF 10 GBPS OR DARK FIBER CONNECTIVITY. THIS ROUTE IS DESIGNED TO MINIMIZE THE NUMBER OF MILES REQUIRED TO MAXIMIZE THE NUMBER OF RURAL COMMUNITIES SERVED. THE ROUTE HAS A HIGH LEVEL OF TOWER AND CUSTOMER DEMAND WHICH WOULD FACILITATE ECONOMIC DEVELOPMENT FOR LOCAL COMMUNITIES, TRIBES, AND THE STATE. THIS MISMATCH IS NOT ONLY OF CONCERN TO RESEARCHERS; FEDERAL AND STATE WILDLIFE AGENCIES ARE ALSO TRYING TO PREPARE FOR THE DOWNSTREAM CONSEQUENCES OF THIS PHENOMENON. ACCORDINGLY, THIS PROJECT WAS DESIGNED IN COLLABORATION WITH WILDLIFE AGENCIES. RESULTS WILL HELP POLICY MAKERS ASSESS EXISTING PROTECTION POLICIES AND TEST NEW MANAGEMENT PLANS FOR MIGRATORY SONGBIRDS. LOCAL OUTREACH ALONG THE MIGRATORY FLYWAY IS ALSO PLANNED (E.G., EDUCATIONAL PROGRAMMING ON MIGRATION THAT WILL CONNECT CHILDREN ACROSS NORTH AND SOUTH AMERICA). UNDERREPRESENTED GROUPS IN STEM WILL ALSO BE RECRUITED IN THIS PROJECT AND INCLUDED IN BOTH THE RESEARCH AND OUTREACH PLANS. PLASTICITY AND ADAPTATION COULD ALLOW MIGRANTS TO ADVANCE THEIR MIGRATORY TIMING, BUT DIRECT ESTIMATES OF THESE MECHANISMS AND THEIR MOLECULAR BASIS ARE RARE AND RESTRICTED TO A SMALL NUMBER OF PROXIMATE POPULATIONS. THIS PROJECT WILL BEGIN FILLING THIS KNOWLEDGE GAP, USING STATE-OF-THE-ART TOOLS FROM SEVERAL DISCIPLINES (E.G., ANIMAL MOVEMENT ECOLOGY, GENOMICS AND COMPUTER MODELING) AND DATA FROM A BROAD GEOGRAPHIC SCALE. SPECIFICALLY, (1) PLASTICITY AND (2) ADAPTATION FOR MIGRATORY TIMING WILL BE ESTIMATED AND (3) THE MOLECULAR BASIS OF THIS TRAIT WILL BE EXAMINED. ESTIMATES OF PLASTICITY AND ADAPTATION WILL THEN BE (4) INTEGRATED INTO MODELS PREDICTING HOW POPULATIONS WILL RESPOND TO CLIMATE CHANGE AND SPECIFIC CONSERVATION STRATEGIES BEING PROPOSED FOR THIS GROUP. HOWEVER, A RECENT COMMUNITY HEALTH NEEDS ASSESSMENT SHOWS THAT THE NEED IS STILL HIGH, AND WAIT TIMES FOR LOCAL PROVIDERS CAN BE 4 WEEKS OR MORE. SMC PLANS TO USE BHSE FUNDING TO HIRE LICENSED BH STAFF, INCREASE MOUD PROGRAM OUTREACH, EXPAND BH SPACE IN CLINIC, AND INVEST IN TRAINING AND TECHNOLOGY TO INCREASE STAFF SKILLS AND EFFICIENCY. REFINING TRICHOMONAS VAGINALIS TREATMENT IN WOMEN AND MEN. - ABSTRACT TRICHOMONAS VAGINALIS IS ESTIMATED TO BE THE MOST COMMON NON-VIRAL SEXUALLY TRANSMITTED INFECTION (STI). IT CAUSES CONSIDERABLE AND COSTLY PERINATAL/REPRODUCTIVE MORBIDITY, DISPROPORTIONATELY AFFECTS PERSONS OF COLOR, AND CAN AMPLIFY HIV TRANSMISSION. AFTER OUR TWO TRIALS FOUND MULTI-DOSE ORAL METRONIDAZOLE (MTZ) TO BE SUPERIOR TO SINGLE-DOSE 2 G ORAL MTZ, THE CENTERS FOR DISEASE CONTROL AND PREVENTION NOW RECOMMENDS MULTI-DOSE MTZ AS THE TREATMENT OF CHOICE FOR T. VAGINALIS IN ALL WOMEN. SINCE NEITHER TRIAL INCLUDED MEN AND THERE IS A PAUCITY OF DATA IN MEN, SINGLE-DOSE 2 G ORAL MTZ STANDS AS THE RECOMMENDED TREATMENT FOR MEN. IN BOTH FEMALE TRIALS, HOWEVER, EVEN THOUGH MULTI-DOSE ORAL MTZ WAS FOUND TO BE SUPERIOR TO SINGLE-DOSE MTZ, MULTI- DOSE MTZ STILL HAD UNACCEPTABLY HIGH RATES OF BREAKTHROUGH INFECTION (9%-11%). WITH APPROXIMATELY 2.6 MILLION CASES OF T. VAGINALIS PER YEAR IN THE UNITED STATES, OVER 280,000 PERSONS/YEAR ARE ESTIMATED TO BE INSUFFICIENTLY TREATED WITH MULTI-DOSE ORAL MTZ. THUS, THERE IS A CRITICAL NEED TO REFINE T. VAGINALIS TREATMENT. SINGLE-DOSE 2 G ORAL SECNIDAZOLE (SEC), A NEXT GENERATION 5-NITROIMIDAZOLE, MAY BE A GOOD OPTION. OUR RECENT TRIAL FOUND THAT SINGLE-DOSE 2 G ORAL SEC WAS SUPERIOR TO PLACEBO IN T. VAGINALIS-INFECTED WOMEN. ORAL SEC HAS MULTIPLE BENEFITS COMPARED TO MULTI-DOSE ORAL MTZ INCLUDING A LONGER HALF-LIFE AND IMPROVED TOLERABILITY. SINGLE-DOSE SEC POSES LESS BURDEN ON THE PATIENT AND CAN TREAT BACTERIAL VAGINOSIS, A COMMON COMORBIDITY AMONG WOMEN WITH T. VAGINALIS. ON THE OTHER HAND, MTZ IS FAR LESS COSTLY AND CAN BE GIVEN DURING PREGNANCY/LACTATION. THE OVERALL GOALS OF THIS MULTI-CENTERED, RANDOMIZED TRIAL ARE TO EXAMINE THE EFFECTIVENESS AND COST-EFFECTIVENESS OF ORAL MULTI-DOSE MTZ COMPARED TO SINGLE-DOSE ORAL SEC INBOTH WOMEN AND MEN WITH T. VAGINALIS. AIM 1. TO EXAMINE THE OPTIMAL TREATMENT FOR T. VAGINALIS INFECTION IN WOMEN AND MEN. THIS WILL BE DONE BY CONDUCTING AN OPEN-LABEL, RANDOMIZED, MULTI-CENTERED, PARALLEL PHASE IV CLINICAL TRIAL COMPARING ORAL MULTI-DOSE MTZ (500 MG BID FOR 7 DAYS) TO SINGLE-DOSE ORAL 2 G SEC FOR THE TREATMENT OF T. VAGINALIS IN WOMEN AND MEN. THE TEST-OF-CURE (TOC) VISIT WILL BE 4 WEEKS (± 1 WEEK) AFTER COMPLETION OF TREATMENT. WE HYPOTHESIZE THAT T. VAGINALIS REPEAT INFECTION RATES AT TOC WILL BE 1.75 LOWER IN THE SINGLE-DOSE 2 G ORAL SEC ARM VERSUS THE MULTI- DOSE ORAL MTZ ARM. VAGINALIS INFECTION AS WELL AS INCREASED RISK FOR HIV-ACQUISITION. WE HYPOTHESIZE THE SINGLE-DOSE SEC WILL HAVE HIGHER INITIAL COST BUT WILL BE MORE COST EFFECTIVE COMPARED TO MULTI-DOSE MTZ, LARGELY DUE TO LOWER BREAKTHROUGH RATES. LOSS OF CORNEAL ENDOTHELIAL CELLS AND GUTTAE (BUMPS IN DESCEMET’S MEMBRANE) ARE PATHOLOGICAL FECD FEATURES. A DEFICIENCY OF CORNEAL ENDOTHELIAL CELLS (CEC) CAUSES CORNEAL OPACITY DUE TO LOSS OF PUMPING FUNCTION. IN DEVELOPED COUNTRIES, INCLUDING THE U.S., FECD PREVALENCE IS ESTIMATED TO BE 4%. CORNEAL TRANSPLANTATION IS THE ONLY TREATMENT FOR ADVANCED FECD PATIENTS. RECENT GENOMIC ANALYSES OF PATIENTS’ DNA HAVE DEMONSTRATED EXPANSION OF (CTG)N TRINUCLEOTIDE REPEATS IN THE INTRON OF THE TCF4 GENE. IN CAUCASIAN POPULATIONS, NEARLY 80% OF LATE-ONSET FECD PATIENTS HAVE LONG (CTG)N (N>30-40) REPEATS, AND THE LENGTH OF (CTG)N IS THE MOST PREDICTABLE GENETIC MUTATION FOR LATE-ONSET FECD. NEVERTHELESS, THE MECHANISM OF HOW THE LONG (CTG)N REPEATS LEAD TO FECD IS UNKNOWN. ONE HYPOTHESIS IS THAT LONG (CUG)N RNA SEQUESTERS RNA SPLICING FACTORS THAT LEAD TO ALTERATION OF RNA SPLICING AND MALFUNCTION OF CECS. AN AVERAGE OF 110 UNDUPLICATED CLIENTS WILL BE SERVED PER YEAR WITH A TOTAL OF 550 SERVED OVER THE LIFETIME OF THE PROJECT. AN MHEALTH APPROACH TO DECREASING OPIOID ABUSE AND OTHER DISEASES OF DESPAIR IN RURAL POPULATIONS - PROJECT SUMMARY/ABSTRACT RURAL AMERICANS FACE NUMEROUS HEALTH DISPARITIES COMPARED TO THEIR URBAN COUNTERPARTS AND ARE MORE LIKELY TO DIE FROM CHRONIC HEALTH CONDITIONS, INCLUDING SUBSTANCE ABUSE. INDEED, OPIOID-RELATED MORTALITY IS HIGHEST IN CERTAIN RURAL AREAS OF THE U.S., INCLUDING APPALACHIA, NEW ENGLAND, AND THE MOUNTAIN WEST. A NUMBER OF STUDIES LOOKING AT MORBIDITY AND MORTALITY PROVIDE INSIGHT INTO THE HEALTH PROBLEMS AFFLICTING LESS EDUCATED, RURAL POPULATIONS.6-9,14-15 LACK OF HIGH SCHOOL DIPLOMA, MENTAL ILLNESS, ALCOHOL USE, AND ILLICIT DRUG USE WERE ALL SUBSTANTIALLY HIGHER IN THE POPULATION OF DECEDENTS, AND INDIVIDUALS SUFFERING FROM THESE PROBLEMS ARE AT HEIGHTENED RISK FOR OPIOID MISUSE.22-29 THE PURPOSE OF THIS PROJECT IS TO DEVELOP A MOBILE-BASED OPIOID PREVENTION PROGRAM FOR RURAL-BASED BLUE-COLLAR WORKERS THAT PROVIDES ROBUST PREVENTIVE INTERVENTIONS AIMED AT REDUCTION IN ALCOHOL CONSUMPTION, MANAGEMENT OF NEGATIVE MOOD STATES INCLUDING ANXIETY AND DEPRESSION, AND NON-PHARMACOLOGIC APPROACHES TO MANAGING PAIN. THE PROGRAM AIMS TO FILL AN IMPORTANT GAP IN SERVICE PROVISION TO RURAL ADULTS AT RISK FOR OPIOID MISUSE. THE PROGRAM WILL CAPITALIZE ON TECHNOLOGIES AVAILABLE IN RURAL COMMUNITIES – NAMELY MOBILE HEALTH – AND WILL BE MEDIA RICH, INCLUDING A PROGRAM ‘COACH’ AND VIDEOS OF INDIVIDUALS THAT USERS CAN RELATE TO DESCRIBING THEIR EXPERIENCES DEALING WITH PAIN, SUBSTANCE USE DIFFICULTIES, STRESS AND MOOD ISSUES - AND THE IMPACT OF THESE ISSUES ON THEIR LIVES. SIMILAR TO OTHER HEALTH PROMOTION AND SUBSTANCE ABUSE PREVENTION PROGRAMS DEVELOPED BY OUR GROUP, THE PROGRAM WILL BE HIGHLY INDIVIDUALIZED - TAILORED BY USER CHARACTERISTICS AND EXPERIENCES, DIRECTING USERS TO MANAGEMENT OF DIFFICULT MOODS, TECHNIQUES IN THE SELF-MANAGEMENT OF PAIN, AND AVOIDANCE OF ABUSING ALCOHOL AND OTHER SUBSTANCES, INCLUDING OPIOIDS. IN PHASE I, WITH CRUCIAL INPUT FROM TWO FOCUS GROUPS AND OUR EXPERT CONSULTANTS, ISA STAFF WILL DEVELOP AND PILOT TEST A MOBILE OPIOID MISUSE PREVENTION PROGRAM EMBEDDED WITHIN A LARGER HEALTH PROMOTION PROGRAM. PRE-DEVELOPMENT FOCUS GROUPS WILL BE CONDUCTED WITH INDIVIDUALS WHO WILL BE RECRUITED TO HELP SHAPE THE STRUCTURE AND CONTENT OF THE PROGRAM. THE FINANCIAL IMPACTS OF HLB ARE STAGGERING AND MAY RESULT IN CITRUS BEING REPLACED BY OTHER FRUIT TREES AND ALTERNATIVE CROPS. IN FLORIDA, WHERE THE DISEASE HAS BEEN WELL ESTABLISHED FOR OVER TEN YEARS, THE YIELD OF CITRUS TREES HAS BEEN REDUCED TO YIELDS REFLECTING THE FLORIDA INDUSTRY 70-100 YEARS AGO. THE REDUCED YIELD IS REFLECTED IN THE HIGH PRICE OF ORANGE JUICE FOR THE MORNING BREAKFAST. THE DISEASE THREATENS THE CITRUS CROP IN OTHER CITRUS-GROWING STATES LIKE CALIFORNIA, TEXAS, AND ARIZONA. FINDING LONG-TERM, SUSTAINABLE SOLUTIONS IS IMPERATIVE FOR CONTINUED CITRICULTURE.CITRUS HLB IS A COMPLEX DISEASE ASSOCIATED WITH A NON-CULTIVABLE BACTERIAL PATHOGEN, SPREAD BY A PSYLLID INSECT VECTOR THAT IS DIFFICULT TO CONTROL, AND THE DISEASE HAS NO KNOWN CURE; INFECTED TREES DECLINE, PRODUCE UNMARKETABLE FRUIT, AND DIE IN A FEW YEARS. MANAGEMENT OF THE DISEASE BY PREVENTING SPREAD, CONTROL OF THE INSECT VECTOR, AND APPLICATION OF ANTI-MICROBIAL SUBSTANCES ARE GOOD PRACTICES - BUT OF LIMITED VALUE FOR A TREE CROP THAT CAN LIVE FOR SEVERAL DECADES. DESPITE INTENSIVE RESEARCH FOR THE PAST FIFTEEN YEARS, TANGIBLE SOLUTIONS TO CONTROL HLB ARE NOT YET AVAILABLE. NOVEL APPROACHES ARE NEEDED TO FIND PRACTICAL METHODS TO MANAGE HLB IN THE CITRUS-GROWING STATES OF THE USA. CURRENT METHODS OF DISEASE MANAGEMENT ARE NOT FINANCIALLY AFFORDABLE. EXCESSIVE USE OF PESTICIDES TO CONTROL THE PSYLLID POPULATIONS IS ENVIRONMENTALLY UNSAFE; PESTICIDES WILL LOSE EFFECTIVENESS AFTER PROLONGED USE. ECOLOGICALLY FRIENDLY BIOLOGICAL CONTROL MECHANISMS THAT REDUCE THE PSYLLID POPULATION WILL NOT BE ADEQUATE TO MANAGE HLB SINCE A SINGLE INSECT VECTOR CARRYING THE PATHOGEN CAN SPREAD THE DISEASE IN A CITRUS GROVE. WHEN THE CITRUS PROCESSING PLANTS AND JUICE FACTORIES ARE CLOSED BECAUSE OF LOW YIELD AND DISEASED FRUIT, IT WILL BE VERY DIFFICULT TO REVIVE THIS INDUSTRY INFRASTRUCTURE. LONG-TERM STRATEGIES FOR DISEASE MANAGEMENT ARE URGENTLY NEEDED TO CONTINUE CITRUS CULTIVATION IN AREAS WHERE THE INSECT VECTOR AND THE PATHOGEN HAVE BEEN ESTABLISHED.CITRUS IS PROPAGATED MAINLY THROUGH GRAFTING. THE VEGETATIVE MODE OF PROPAGATION OF A LIMITED NUMBER OF CULTIVARS HAS RESULTED IN INNUMERABLE CULTIVATED TYPES, BUT THERE IS LITTLE GENETIC DIVERSITY IN CULTIVATED CITRUS. WHEN NEW PATHOGENS ARRIVE, THIS GENETIC UNIFORMITY CAN RESULT IN DISEASE EPIDEMICS AND DIRE CONSEQUENCES FOR THE CROP. IN ADDITION, THERE IS NO DOCUMENTED RESISTANCE TO HLB IN CULTIVATED CITRUS. HOWEVER, OTHER CITRUS-RELATED WILD PLANTS MAY HAVE RESISTANCE TRAITS TO HLB. BASED ON OUR PREVIOUS WORK, WE DETERMINED THAT CERTAIN NON-CITRUS TAXA THAT ARE CLOSELY RELATED TO CITRUS HAVE DISEASE-RESISTANCE TRAITS AND THESE MAY BE UTILIZED TO IMPART THE MUCH-NEEDED HLB RESISTANCE TO CULTIVATED CITRUS TYPES. OUR APPROACH IS TO GENERATE NOVEL HYBRIDS OF CITRUS USING CONVENTIONAL BREEDING METHODS WITH SEXUALLY C,OMPATIBLE CITRUS RELATIVES HAVING GENETIC RESISTANCE TO THE HLB PATHOGEN. TOWARDS THIS GOAL, WE HAVE CONDUCTED A BREEDING PROGRAM SINCE 2013 BY CROSSING CITRUS ACCESSIONS WITH DISEASE-RESISTANT/TOLERANT AUSTRALIAN LIME SPECIES BELONGING TO THE GENUS MICROCITRUS. CROP RELATIVES ARE UTILIZED AS SOURCES OF BENEFICIAL TRAITS IN OTHER CULTIVATED PLANTS. CITRUS IS SEXUALLY COMPATIBLE WITH MANY CLOSELY RELATED GENERA; THIS BIOLOGICAL PECULIARITY OF CULTIVATED CITRUS TYPES WAS EXPLOITED IN OUR BREEDING PROGRAM TO GENERATE HUNDREDS OF INTERGENERIC HYBRIDS.CITRUS BREEDING IS CONSIDERED VERY DIFFICULT AND TIME-CONSUMING SINCE THE GENERATION TIME CAN BE 4 TO 5 YEARS. SINCE OUR BREEDING PROGRAM HAS BEEN IN PROGRESS FOR TEN YEARS NOW, WE HAVE GENERATED PROGENY FROM SEVERAL DIFFERENT TYPES OF CROSSES. EVALUATION OF THE NOVEL HYBRIDS IN THE FIELD AND IN THE GREENHOUSE DETERMINED THAT HLB RESISTANCE TRAITS CAN BE INHERITED BY A SMALL PROPORTION OF THE BREEDING PROGENY. HOWEVER, SINCE ONE PARENT OF THE CROSS BEARS FRUIT THAT IS BARELY EDIBLE, MANY HYBRIDS OF THE FIRST-GENERATION CROSSES HAVE FRUITS THAT MAY NOT BE ACCEPTABLE AS SUBSTITUTE VARIETIES, EVEN THOUGH THEY HAVE DISEASE-RESISTANCE TRAITS. WE HAVE NOW GENERATED HYBRIDS OF THE SECOND GENERATION BY CROSSING THE PROMISING HYBRIDS FROM THE FIRST GENERATION TO COMMERCIAL TYPES OF CITRUS. A SMALL PERCENTAGE OF THE ADVANCED HYBRIDS IS EXPECTED TO HAVE RETAINED THE HLB RESISTANCE TRAITS AND MAY HAVE FRUIT QUALITIES SIMILAR TO COMMERCIAL CITRUS TYPES. WE PROPOSE TO UTILIZE MOLECULAR APPROACHES TO DEVELOP MARKERS THAT WILL AID IN THE SELECTION OF THE BREEDING PROGENY FROM THE SECOND GENERATION OF HYBRIDS TO IDENTIFY INDIVIDUALS WITH PROMISING HLB RESISTANCE. IF SUCH HYBRIDS CAN RESIST HLB IN FIELD CONDITIONS AND IF THEY HAVE FRUIT TRAITS THAT ARE DEEMED ACCEPTABLE BY THE PUBLIC, WE WILL HAVE NOVEL HYBRIDS WITH DISEASE RESISTANCE AND ACCEPTABLE ORGANOLEPTIC ATTRIBUTES. CULTIVAR DEVELOPMENT OF PROMISING INDIVIDUALS WILL BE REQUIRED BEFORE LARGE-SCALE CULTIVATION CAN BE ADOPTED.IF NOVEL HYBRIDS WITH GENETIC RESISTANCE TO HLB ARE DEVELOPED, IT WILL BE POSSIBLE TO CULTIVATE CITRUS DESPITE HAVING THE PATHOGEN AND THE INSECT VECTOR IN THE AREA. MANAGEMENT OF THE GROVES THAT HAVE RESISTANT CULTIVARS WILL BE SIMILAR TO THE GROVE MANAGEMENT DONE BEFORE HLB ARRIVED. FINANCIALLY IT WILL BE FEASIBLE TO CULTIVATE CITRUS IF ELABORATE, EXPENSIVE MANAGEMENT PRACTICES ARE NOT NECESSARY. ANOTHER ADVANTAGE IS THAT THE NOVEL HYBRIDS MAY HAVE NEW FLAVORS THAT MAY ENHANCE THE KNOWN CITRUS FLAVORS THAT ARE GENERALLY ASSOCIATED WITH CITRUS FRUITS. WE WILL LOOK FOR THE METABOLOMICS PROFILE OF NOVEL HYBRIDS TO IDENTIFY THE FLAVOR-ENHANCING COMPOUNDS IN THE HYBRID FRUITS AND COMPARE THEM WITH STANDARD CITRUS. REPLACEMENT OF PRESENTLY CULTIVATED CITRUS VARIETIES WITH NOVEL HYBRIDS WILL BE BENEFICIAL TO MANAGE HLB. CONSTRUCTION OF 579 MILES FIBER ROUTES AND USE OF 127 MILES OF FIBER IN APPALACHIAN POWER COMPANY'S (APCO) EXISTING NETWORK. 2. PROVIDE EFFICIENT AND COST-EFFECTIVE ACCESS AT INTERCONNECTION POINTS TO INTERNET SERVICE PROVIDERS (ISPS), ENABLING RELIABLE AND AFFORDABLE BROADBAND ACCESS TO APPROXIMATELY 113,000 RESIDENTS. 3. FACILITATE THE PROVISION OF BROADBAND SERVICE AT SPEEDS NOT LESS THAN 1 GBPS SYMMETRICAL SERVICE FOR ANCHOR INSTITUTIONS WITHIN 1,000 FEET OF THE PROPOSED PROJECT. 4. THIS PROJECT IS PROJECTED TO IMPACT 2,955 UNSERVED HOUSEHOLD AND 20,676 UNDERSERVED HOUSEHOLD AND THESE NUMBERS ARE LIKELY UNDERSTATED PER THIRD-PARTY DATA. BASED ON ECONOMIC ANALYSES, IN CONSERVATIVE ESTIMATES, INCREASED BROADBAND CONNECTIVITY WOULD RESULT IN AN ECONOMIC UPLIFT, AN UNEMPLOYMENT RATE REDUCTION, AND CREATE 180 NEW JOBS ANNUALLY.
