FURTHERMORE, THESE SATELLITES GENERATE FLARES AND GLINTS THAT CAN EASILY BE CONFUSED WITH REAL ASTROPHYSICAL EVENTS, ULTIMATELY RESULTING IN BOGUS TRANSIENT ALERTS AND ARTIFICIAL FAINT GALAXIES. CONSEQUENTLY, THE NEED FOR MITIGATION IS CRITICAL, AND THE TIMESCALE FOR DEVELOPMENT IS SHORT. THIS PROJECT WILL USE ARTIFICIAL INTELLIGENCE (AI) TO DEVELOP TECHNIQUES OF ADDRESSING THESE CHALLENGES. THE PROJECT WILL LEVERAGE STATE-OF-THE-ART AI TO MITIGATE THE SYSTEMATIC ERRORS THAT DEGRADE IMAGE QUALITY AND THREATEN THE SCIENTIFIC IMPACT OF THE LSST. WORKING IN CLOSE COLLABORATION WITH ASTRONOMERS, SPACEX, AND OTHER CONSTELLATION OPERATORS, THE INITIATIVE UTILIZES GOOGLE DEEPMIND GEMINI 3.5 PRO AS ITS CORE AI ENGINE. TRAINED ON LAB IMAGES OF SATELLITE STREAKS ON A REALISTIC SKY, A NOVEL AI STREAK DETECTOR ALREADY PRODUCES HIGH ACCURACY AND COMPLETENESS. HOWEVER, THIS PROJECT WILL IMPROVE THE PERFORMANCE. FIRST, USING RUBIN LSSTCAM IMAGING DATA THE AI WILL GENERATE A COMPREHENSIVE DATABASE CATALOGING THE NUMBER, ORIENTATION, FLUX PROFILE, AND LOCATION OF STREAKS ON EACH IMAGE. SECOND, THE ENGINE WILL BE FURTHER TRAINED ON A SPECIALIZED HARDWARE SIMULATOR OF LSST SATELLITE STREAKS TO DRASTICALLY IMPROVE THE ACCURACY AND COMPLETENESS OF AUTOMATED DETECTION. FINALLY, THE PROJECT WILL DEVELOP ADVANCED CHARACTERIZATIONS OF THESE STREAKS TO ASSIST RESEARCHERS WITH SYSTEMATIC ERROR ANALYSIS AND DATA MASKING, AND SUPPORT SATELLITE OPERATORS IN THEIR REFLECTIVITY MITIGATION EXPERIMENTS. ULTIMATELY, THIS WORK WILL DELIVER CALIBRATED MONITORING FOR A SATELLITE POPULATION APPROACHING ONE MILLION, PROVIDING THE GLOBAL ASTRONOMY COMMUNITY WITH AN INCREASED UNDERSTANDING OF SYSTEMATIC ERRORS AND VASTLY IMPROVED DATA RELIABILITY. DECIPHERING THE ECOLOGICAL AND EVOLUTIONARY RULES OF FUNGAL HORIZONTAL GENE TRANSFER -HORIZONTAL GENE TRANSFER (HGT) IS THE TRANSFER OF GENETIC MATERIAL BETWEEN ORGANISMS THAT ARE NOT PARENT AND OFFSPRING, AND MAY EVEN BE FROM DIFFERENT SPECIES. UNLIKE TRAITS PASSED FROM PARENT TO OFFSPRING, HGT LETS ORGANISMS GAIN NEW TRAITS THROUGH CONTACT OR PROXIMITY WITH ANOTHER INDIVIDUAL. MANY EXAMPLES EXIST IN NATURE, BUT THE ROLE OF HGT IN EUKARYOTES IS STILL DEBATED. IN MANY CASES, THERE IS CLEAR EVIDENCE THAT HGT HAS OCCURRED, BUT FEW KNOWN MECHANISMS TO EXPLAIN HOW OR WHY IT HAPPENS. THIS PROJECT BUILDS ON A RECENT DISCOVERY OF A CLEAR HGT MECHANISM IN FUNGI, A MAJOR GROUP OF EUKARYOTES, TO ANSWER KEY QUESTIONS ABOUT HOW THIS PROCESS WORKS. THE WORK WILL IMPROVE UNDERSTANDING OF FUNGI THAT AFFECT HUMAN, PLANT, AND ENVIRONMENTAL HEALTH. IT MAY ALSO SUPPORT NEW ADVANCES IN EVOLUTION AND BIOTECHNOLOGY, SINCE GENE TRANSFER BETWEEN EUKARYOTIC SPECIES COULD HELP SOLVE PROBLEMS IN PLANT BREEDING, MEDICINE, AND DRUG DISCOVERY. IN ADDITION, THIS PROJECT WILL HELP BUILD THE WORKFORCE BY TRAINING EARLY-CAREER RESEARCHERS, SUPPORTING TEAM-BASED STUDENT LEARNING, AND EXPANDING MENTORING PROGRAMS. UNDERSTANDING HOW SPECIES EXCHANGE GENETIC INFORMATION THROUGH HGT IS CRITICAL FOR UNDERSTANDING THE GENETIC BASES OF ADAPTATION. YET DESPITE ITS IMPORTANCE FOR GENERATING ADAPTIVE VARIATION, HGT IN EUKARYOTES HAS BEEN NEAR-IMPOSSIBLE TO STUDY BECAUSE THE FIELD LACKS A SYSTEM FOR HYPOTHESIS-DRIVEN EXPERIMENTATION. THIS PROJECT ADDRESSES THIS KNOWLEDGE GAP BY CAPITALIZING ON THE PI?S ONGOING WORK, WHICH REVEALS THAT HGT WITHIN FUNGAL EUKARYOTES IS MEDIATED BY GIANT TRANSPOSONS CALLED STARSHIPS CAPABLE OF TRANSFERRING BETWEEN SPECIES UNDER LAB CONDITIONS AND IN NATURE. THE OVERARCHING GOAL OF THIS PROJECT IS TO DEVELOP STARSHIPS INTO A MODEL SYSTEM TO EXPERIMENTALLY DETERMINE THE ECOLOGICAL AND EVOLUTIONARY FACTORS AFFECTING FUNGAL HGT. IN DOING SO, THE PI WILL ADDRESS OUTSTANDING QUESTIONS ABOUT THE MODE AND TEMPO OF EUKARYOTIC ADAPTATION WITH A LONG-TERM GOAL TO LEVERAGE KNOWLEDGE OF FUNGAL HGT TO DEVELOP CUSTOM TRANSFORMATION-BASED BIOTECHNOLOGIES FOR EUKARYOTIC SYSTEMS. THIS PROJECT ADDRESSES THREE INDEPENDENT AND COMPLEMENTARY AIMS USING A NOVEL HGT ASSAY: 1) TO DETERMINE HOW RATES OF HGT CHANGE IN RESPONSE TO DIVERSE ENVIRONMENTAL CUES; 2) TO INVESTIGATE WITHIN-SPECIES VARIATION IN HGT TRANSMITTER AND RECEIVER ABILITIES AND DETERMINE THE GENETIC BASES OF THESE TRAITS; 3) TO TEST IF CLOSER RELATIVES WITHIN A MODEL GENUS TRANSFER GENES MORE OFTEN AND WHETHER MORE PHYLOGENETICALLY VARIABLE COMMUNITIES FACILITATE MORE DISTANT TRANSFERS. SPECTRAL FINGERPRINTS ENCODED WITHIN THAT LIGHT REVEAL THE TEMPERATURES, CHEMICAL COMPOSITION, DENSITY, AND MOTIONS OF OBJECTS THROUGHOUT THE COSMOS. TURNING THESE OBSERVATIONS INTO SCIENTIFIC DISCOVERIES REQUIRES SOPHISTICATED COMPUTER MODELS THAT CONNECT THE OBSERVED LIGHT WITH THE PHYSICAL CONDITIONS THAT PRODUCED IT. YET THE UNDERLYING MOLECULAR MECHANISMS DRIVING STARSHIP-MEDIATED VARIATION IN FUNGAL POPULATIONS AND THEIR EVOLUTIONARY CONSEQUENCES REMAIN UNKNOWN. HERE, WE WILL DETERMINE THE EVOLUTIONARY MODES AND TRANSMISSION ROUTES OF STARSHIPS USING THE MODEL ORGANISM AND OPPORTUNISTIC FUNGAL PATHOGEN ASPERGILLUS FUMIGATUS. IN AIM 1, WE WILL DETERMINE IF STARSHIP-ASSOCIATED PARALOGS GENERATE MORE DIVERGENT PHENOTYPIC VARIATION COMPARED WITH THEIR GENOMIC PARALOGS. WE BUILD ON THE STRENGTHS OF OUR KL2 PROGRAM, WHICH HAS TRAINED 27 SCHOLARS SINCE ITS INCEPTION IN 2010. COLLECTIVELY, THEY HAVE PUBLISHED 978 PEER-REVIEWED PUBLICATIONS AND GARNERED $339 MILLION AS PIS IN EXTRAMURAL FUNDING, WITH ALL REMAINING IN RESEARCH-INTENSIVE CAREERS. GOAL AND RATIONALE: THE OVERALL GOAL OF OUR NEW K12 PROGRAM IS TO DEVELOP THE CAREERS OF SCIENTISTS WHO WILL DRIVE THE AGENDA OF ACCELERATING THE TRANSLATIONAL PROCESS AND CHANGING PARADIGMS IN TRANSLATIONAL SCIENCE (TS). THIS PROJECT WILL FURTHER HELP SCIENTISTS AND LAND MANAGERS MAKE BETTER FORECASTS FOR THE KINDS OF TRAITS THAT ANIMAL SPECIES WILL NEED IN ORDER TO ESTABLISH POPULATIONS AT HIGHER ELEVATIONS. WITHOUT TREATMENT, SMM CAN LEAD TO PREVENTABLE MATERNAL DEATHS. INTERVENTIONS TO REDUCE SMM DISPARITIES HAVE LACKED SPECIFIC MECHANISMS TO REACH AT-RISK WOMEN WITHIN COMMUNITIES EXPERIENCING HEIGHTENED SMM RISKS. COMMUNITY-BASED PATIENT NAVIGATION IS AN EMERGING SUCCESSFUL APPROACH TO IMPROVING COMMUNITY HEALTH. WE PROPOSE A COMMUNITY-CENTRIC INTERVENTION, CALLED PATHWAYS THROUGH PREGNANCY (PTP), WITH THE GOAL OF DECREASING SMM THROUGH OUTREACH BY COMMUNITY-BASED HEALTH NAVIGATORS THAT HELP PREGNANT WOMEN IDENTIFY AND SURMOUNT BARRIERS-TO-CARE THAT LEAD TO SMM RISK. PTP NAVIGATORS WILL BE SUPPORTED BY A SMARTPHONE APP CALLED PATHWAYS. THREE PHASE I AIMS ARE PROPOSED: AIM 1) DESIGN THE PATHWAYS THROUGH PREGNANCY (PTP) INTERVENTION WITH INPUT FROM PREGNANT AND RECENTLY PREGNANT WOMEN, COMMUNITY MEMBERS, AND HEALTHCARE PERSONNEL THROUGH A USER-CENTERED, PARTICIPATORY DESIGN APPROACH, USING DATA FROM A LOW-INCOME STUDY COMMUNITY IN BALTIMORE; AIM 2) DESIGN AND PROTOTYPE THE PATHWAYS APP, DRAWING ON MULTIPLE STAKEHOLDER PERSPECTIVES AND USING A USER-CENTRIC PARTICIPATORY PROCESS TO PRODUCE AN INTERACTIVE PATHWAYS PROTOTYPE; AIM 3) CONDUCT A MULTI-PERSPECTIVE ASSESSMENT OF PTP AND THE PATHWAYS APP FOR FEASIBILITY FOR FURTHER DEVELOPMENT, TRANSLATION, AND COMMERCIALIZATION, USING BOTH QUALITATIVE AND QUANTITATIVE METHODS AND CRITERIA OF PERCEIVED USEFULNESS OF PTP OVERALL AND USABILITY OF PATHWAYS.
